Yes, there is a perfect spot for rupee farming. As long as you kill everything on every screen along the way. The enemies won't respawn for a while if some remain, which defeats the purpose. It will be an endless line of easy-to-kill high-yield enemies.

First, warp to Magical Shortcut, located east of Kakariko Village in the far south of Hyrule. This was the best place to farm rupees in Zelda 1. Of course it had to remain that way. Kill all the blue tektites in the larger area as you head east. You'll get to some red leevers. Keep going east and you'll enter the Southern Shoreline. Go east a little further, through some red leevers and red octorocs, and into the warp. Use the world map to go back to Magical Shortcut. Enough time has passed that every enemy will be back. Loop as many times as needed.

This is the best path to take to get all the money-dropping blue tektites and red leevers. The Magical Shortcut warp is on the left, and the warp in Southern Shoreline is on the right.
